Objectives:

1. To establish the course of the project and the timetable and requested acquisitions.

2. The selection of he subjects for the trial and beginning to create the studied groups.

The beginning of subjects exclusion based upon inclusion/exclusion criteria and personal data registration.

The effectuation of the oral glucose tolerance test with 75g glucosis in non diabetics subjects. The recording of height, weight of the patients and the calculus of bod mass index. The beginning of groups creation based upon body mass index and oral glucose tolerance test.

2008

Objectives:

1. The finishing of the studied groups and idenftification of the CNR1 polymorphisms recorded in genetics data base: 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G
The continuing of subjects inclusion in the trial based on inclusion/exclusion criteria and personal data recording. The effectuation oral glucose tolerance test in non diabetics subjects.
The recording of height, weight of the patients and the calculus of bod mass index.
The definitivation of the 4 groups based on body mass index and OGTT.
Genotyping of five polymorphisms of the CNR1 gene: 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G

2. To establish if 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G- polymorphisms of the CNR1 gene confer high risc for the development of IR and type 2 diabetes mellitus

3. The evaluation of the - 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G- polymorphisms of the CNR1 gene in the modulation of the endocannabinoids’s role in the physiopatogeny of the pancreatic beta cell insulin secretion.

Objectives:

1. To establish if 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G- polymorphisms of the CNR1 gene confer high risc for the development of dyslipidemia.
Dosage:

  • total cholesterol
  • HDL-cholesterol
  • Tryglicerides

The calculus of LDL-cholesterol using Friedwald’s equation.
The analysis of the prelimnary data.

2. The evaluation of the correlation of - 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G- polymorphisms with obesity.
The analysis of the anthropometric parameters (weight, waist circumference, hip circmference, body mass index waist-hip-ratio) and the determination of the obesity’s type – abdominal or total.

3. The evaluation of the correlation of - 1359 G>A, 23866A>C, 10908G>A, 3813 A>G, 4894 A>G- polymorphisms with adiponectin and leptin levels.

  • fasting adiponectin levels
  • fasting leptin levels

2010

Objectives:

1. The identification of new polymorphisms of CNR1 gene by scanning a 1000bp of the CNR1 gene.
The scan of a 1000bp region of the CNR1 gene to identify new polymorphisms.

The analysis of the prelimnary data.

2. The evaluation of the correlation of the new polymorphisms of the CNR1 gene with metabolic phenotype and pancreatic insulin secretion.

The analysis of the correlation of the new polymorphisms of the CNR1 gene with:

  • IR
  • Type 2 diabetes mellitus
  • Obesity
  • Dyslipidemia
  • Insulin secretion

The dissemination of the results according to the agreement of the financer.

  • participation at national and international scientific debates
  • creating papers for print in scientific magazines.

Final data analysis.

The presentation of the project’s results and Project’s Final Report.
